Subject

Аim: To determine the drug release from lipid based formulations in an in vitro model of the gastrointestinal tract. Materials and methods: 5 types of surfactants and one type of drug - fenofibrate - were studied. High performance liquid chromatography was used to determine the concentration of the dissolved fenofibrate. Results: Best solubilization was observed in the systems where enzymes were absent. The lipid carrier from which the greatest release of fenofibrate was achieved was Tween 80 (587 +/-36 ug/mL), followed by Peceol (347 +/-80 ug/mL), Sunflower Oil (108 +/-20 ug/mL), Sugar Ester OWA 1570 (72 +/-11 ug/mL) and Lauroglycol FCC (40 +/-8 ug/mL). The only system where higher dissolved drug concentrations were achieved in the presence of enzymes was OWA 1570. Discussion: The type of carrier used and the enzymes have a strong influence on the concentration of dissolved fenofibrate under conditions, resembling those of the gastrointestinal tract. The highest concentrations of fenofibrate were measured using Tween 80 and Peceol based systems. The negative influence of the enzymes for some of the systems is most likely due to the partial hydrolysis of the ester bond of the carrier.
